Abstract

<PICT:0986385/C2/1> The antitumour agents and antibiotics Hilamycins A, B and C are obtained by cultivating a strain of Streptomyces rochei var. hilamyceticus in an aqueous nutrient medium under submerged aerobic conditions, preferably at 25-35 DEG C. Hilamycin A is a white solid or colourless syrup soluble in alcohols, acetone, esters, benzene and in water at alkaline pH, but insoluble in water at acid or neutral pH, petroleum ether and hexane; having a U.V. maximum at 235-240 mm ; having an I.R. spectrum as shown in Fig. 2; giving negative ferric chloride and Tollens reactions; having elemental analysis C=69.22, H=9.74 and (a )25 DEG D=0. Hilamycin B is a colourless syrup, having similar solubilities to Hilamycin A; a U.V. maximum at 200 mm ; I.R. bands at 3360, 3200, 2940, 2880, 1725, 1705, 1660, 1458, 1437, 1375, 1240, 1160, 1100, 1040, 964, 818, 745 and 695 cm.-1; elemental analysis C = 66.24, H = 8.05, O = 22.53 and N = 1.16 and (a )20 DEG D = 63 DEG (1% in methanol). Hilamycin C is a substance which is separated from Hilamycins A and B by its solubility in hexane or petroleum ether and has U.V. maxima at 226 and 258 mm , has I.R. bands at 2910, 2850, 1700, 1473, 1431, 1406, 1347, 1304, 1285, 1265, 1245, 1225, 1206, 1185, 1095, 932 and 712 cm.-1 and is active against bacteria. The antitumour agents Hilamycins A, B and C are purified by (1) extraction of the culture filtrate at an alkaline pH with an organic solvent and addition of a non-solvent thereto, (2) transferring them from aqueous solution to an organic solvent at acid or neutral pH, (3) transferring them from the organic solvent solution to an aqueous solution at weak alkaline pH, (4) precipitating them from aqueous solution by addition of acid or (5) adsorbtion of an alkaline extract of the culture filtrate on a phenol type ion exchange resin. The antitumour agents are separated from each other by (1) passing an organic solvent solution thereof over active carbon, eluting Hilamycin B and then A with methanol-acetone and Hilamycin C with methanol-benzene or (2) counter-current distribution. The antitumour agents are used to treat mouse leukaemia by intravenous injection.
