Abstract

<PICT:1057051/C2/1> An antibiotic tetrahexin, active against yeasts, moulds and gram positive bacteria, is characterized as containing polygenic groups and is soluble in dimethylformamide and in acetic acid, insoluble in conventional organic solvents and water of pH 3-9 but sparingly soluble in C1- 8 alkanols and in water of pH lower than 3 and higher than 9; it is amphoteric; has elementary analysis C=65.55%, H=8.46%, N=1.32% and O=24.67%; U.V. adsorption maxima at 368 mm E1%1 cm. = 440; 350 mm E1%1 cm.=466; 318 mm E1%1 cm.=630; 304 mm E1%1 cm. = 616; 290 mm E1%1 cm. = 420 and shoulder at 335 mm ; Rf values as shown in Table III; I.R. as shown in Fig. 1. Tetrahexin is obtained by cultivating streptomyces tetrahexinicus A.T.C.C. 14972 in an aqueous medium containing a source of carbon, nitrogen and essential inorganic salts, at a pH value of 4.5 to 9 and preferably 7 under aerobic submerged conditions. It may be isolated from the separated mycelium by extraction with a C1- 8 alcohol, followed by purification by counter-current distribution or solution in acetic acid and precipitation by neutralization with NaOH and dilution with water.
