Abstract
Interferon is purified by concentrating an aqueous inorganic salt solution of interferon by pressure dialysis, subjecting the concentrate to chromatography on sulphomethylcellulose at pH 2 with an increasing chloride gradient and subjecting the purified material so obtained to one or more stages of chromatography on diethylaminoethylcellulose at pH of between 5 and 6 with an increasing phosphate gradient and recovering a fraction containing interferon of increased purity and concentration. The six stage purification of the example consists of (a) ammonium sulphate precipitation followed by pressure dialysis, (b) chromatography on sulphomethylcellulose at pH 2,0, (c) concentration and adjustment to pH 6,6, (d) chromatography on DEAE cellulose at pH 6,6, (e) adjustment to pH 4,5 and chromatography on DEAE cellulose at pH 4,5, (f) adjustment to pH 5,8 and chromatography on DEAE cellulose at pH 5,8. Step (d) may be carried out at pH 6,g to 8,0.
