Abstract
<PICT:0975771/C1/1> <PICT:0975771/C1/2> The antibiotic porfiromycin is prepared by cultivating a porfiromycin-producing strain of Streptomyces ardus especially S. ardus NRRL 2817 in an aqueous nutrient medium containing a source of carbon and nitrogen under aerobic preferably submerged aerobic conditions until substantial porfiromycin activity is imparted to said medium. Cultivation may be carried out at 18-40 DEG C. for 2-10 days. The antibiotic is isolated by extracting the culture filtrate with a water-immiscible organic solvent e.g. ethyl acetate. The antibiotic may be purified by (a) countercurrent distribution with water and ethyl acetate (1:1) (b) adsorption on to alumina or carbon and elution. The antibiotic Porfiromycin is effective in inhibiting the growth of gram negative and gram positive bacteria and when isolated in the pure form is characterized by (a) forming dark purple crystals melting at 201-201.5 DEG C. (b) empirical formula C16H20N4O5 (c) a paper strip mobility as shown in Fig. 2. (d) an I.R. spectrum as shown in Fig. I (e) elemental analysis C=55.44 H=6.00 N=15.98 O=23.06 (f) [a ]25D=275 DEG \sB55 (C=0.01% methanol) and [a ]25430= +2525 DEG \sB50 (C=0.02% methanol) (g) U.V. maxima at 217, 248, 364 +575mm and (h) being soluble in water, water-immiscible polar organic solvents + water-miscible organic solvents and relatively insoluble in diethyl ether, toluene and cyclohexane. The antibiotic may be added to the feed and drinking water of animals and fish as a prophylactic.ALSO:Porfiromycin (see Division C2) is fed to animals and birds e.g in their feed or drinking water to promote the growth thereof.ALSO:Aqueous solutions of porfiromycin (see Division C2) are used to disinfect instruments, rooms, clothes, paper and fabric by washing.
