Abstract

The invention comprises compounds of the general formula: <FORM:0969077/C1/1> (wherein X is an ethylene or methyl-substituted ethylene group and S stands for one or two chlorine, bromine or fluorine atoms or a p-nitro group) in their racemic and optically active forms, medicinally acceptable acid addition salts thereof, their preparation by quaternising an isoquinoline of the general formula: <FORM:0969077/C1/2> (wherein S1 stands for one or two chlorine, bromine or fluorine atoms and the broken lines denote optional substituents) with a methylating agent, converting the resulting quaternary salt to the corresponding quaternary ammonium hydroxide, subjecting the latter to a Hofmann degradation to produce an olefinic compound of the formula: <FORM:0969077/C1/3> hydrogenating this compound at the olefinic bond, nitrating the hydrogenated compound when it contains an unsubstituted phenyl group and, if desired, converting the hydrogenated or hydrogenated and nitrated compound as the case may be to an acid addition salt, and pharmaceutical preparations containing a compound of the invention in association with a carrier. The optically active compounds may be prepared by using an optically active starting material or by using a racemic starting material and separating the optical antipodes after the hydrogenation step. 1-(2-Vinyl-4, 5-dimethoxyphenyl) -1- dimethylamino -3- (4-chlorophenyl)- propane is characterized as the methiodide and D-camphorsulphonate. (-) and (+) 1-[(3, 4-Dichlorophenyl)-ethyl]-2-methyl-6, 7-dimethoxy-1, 2, 3, 4-tetrahydroisoquinoline are prepared from the racemic compound. The compounds of the invention possess analgesic, spasmolytic, antitussive and bloodpressure lowering properties and may be administered enterally or parenterally in the form of pharmaceutical preparations (e.g. tablets, dragees, suppositories, capsules, solutions, suspensions or emulsions) containing them, with or without other (unspecified) therapeutically valuable substances, in association with a carrier.
