Abstract
Sterile, injectable therapeutic compositions comprise an injectable liquid carrier containing microscopic, medicament-containing capsules produced by liquid-liquid phase separation. Suitable encapsulating materials include gelable hydrophillic colloids e.g. gelatin, albumin, fibrinogen, casein, agar, sodium oleate, pectin and ichtlyocolla which may be used alone or together with other colloid materials of opposite charge e.g. gum acacia, sodium carboxymethylcellulose, sodium alginate and cellulose-, amylose- and starch acetate phthalates and "linear macromolecular synthetic polymers" i.e. materials whose structure comprises both lipophilic and hydrophilic units e.g. those derived from styrene, and an alkyl ring-substituted styrene or an ether- or ester-substituted ethylene and those derived from maleic, crotonic or acrylic acids or maleic acid or acrylic acid amides respectively. Examples of such polymers are styrene-maleic acid and styrene-maleic acid copolymers, sulphonated polystyrenes, polymethacrylic acid and vinyl ether-maleic acid copolymer. The particles to be encapsulated may be solid, liquid or emulsion particles having either hydrophilic or lipophilic surface characteristics and the encapsulation may be achieved by any suitable liquid/liquid phase separation process e.g. those disclosed in Specifications 751,600, 911,483, 929,402, 929,408 and 929,409. The capsules may be hardened by chemical or physical means e.g. irradiation and their components may be sterilized by conventional techniques e.g. by heat, disinfecting gases or irradiation and the encapsulation process may be carried out under aseptic conditions. Specific examples relate to medicaments comprising solutions of testosterone propionate and 6 - methyl - D 1 - hydrocortisone - 21 - acetate in cottonseed oil encapsulated in gelatin/acacia, and a cottonseed oil-in-water emulsion containing methyl salicylate in the aqueous phase and D 1-hydrocortisone acetate in the oil phase encapsulated in gelatin/styrenemaleic acid copolymer.
