Abstract
The invention comprises compounds of the general formula <FORM:0905343/IV (b)/1> (wherein X represents a hydrogen or halogen atom and R1 and R2 each represent a hydrogen atom or a C1-4 alkyl group), and the preparation thereof by converting an acid of the general formula <FORM:0905343/IV (b)/2> (wherein Alk represents a C1-4 alkyl group) into its acid halide, reacting this with diazomethane to give a diazoketone, and subjecting this to thermal decomposition in benzyl alcohol or cyclohexanol, followed by hydrolysis of the resulting ester, and finally, if desired, converting the alkoxy groups to hydroxy groups by the action of a hydrohalic acid. The products are intermediates for the synthesis of pharmaceutical compounds, especially of the tetracycline series. Acids of the second general formula above are prepared from compounds of the general formula <FORM:0905343/IV (b)/3> (wherein A represents a halogen atom or an alkoxy group) by reducing the group -CO-A to -CH2OH, converting the alcohol into its hydrohalic or p-toluenesulphonic ester, condensing this with a sodium carbethoxysuccinic or -methylmalonic acid dialkyl ester, cyclizing the product by treatment with polyphosphoric acid, hydrolysing any esterified carboxyl groups still present, and partially decarboxylating the resulting dicarboxylic acid.
