Abstract
Hydrocortisone is prepared by culturing an aqueous nutrient medium containing 5-pregnene-3b ,17a ,21-triol-20-one or its 3,21-diacetate or its 3- or 21-acetate with Corynebacterium mediolanum under aerated conditions to produce a cultured broth containing 17a -hydroxy-desoxycorticosterone and treating the resulting broth with a broth of the micro-organism Cunninghamella blakesleeana cultured in a nutrient culture medium. The ratio of broths is preferably between 14 : 1 and 4 : 1 by volume (C. blakesleeana to C. mediolanum). Vitamin K may be added when the broth containing the microorganisms C. mediolanum is mixed with that containing the C. blakesleeana micro-organisms. In addition, a mixture of equal parts of 95% ethanol and acetone by volume may be added to the C. mediolanum broth when it is mixed with the C. blakesleeana broth. Detailed examples are given.
