Abstract

The invention comprises compounds of the general formula: <FORM:0856529/IV(b)/1> and their acid addition and quaternary ammonium salts (wherein X is a hydrogen or halogen atom, a lower alkyl or alkoxy group, or a cyano.methylthio, methanesulphonyl, dimethylsulphamoyl or trifluoromethyl group; R is a hydrogen atom or methyl group; and R1 is a lower alkyl group), and the preparation thereof by: (1) reacting a compound of the general formula: <FORM:0856529/IV(b)/2> (wherein Y is the acid residue of a reactive ester), or an acid addition salt thereof, with a compound of the general formula: <FORM:0856529/IV(b)/3> (2) reacting a compound of the general formula: <FORM:0856529/IV(b)/4> with a compound of the general formula: <FORM:0856529/IV(b)/5> (3) reacting a compound of the general formula: <FORM:0856529/IV(b)/6> with a compound of the general formula: Hal-SO2R1 (wherein Hal is a halogen atom); or (4) decarboxylating a compound of the general formula: <FORM:0856529/IV(b)/7> by heating to a temperature above 100 DEG C. until evolution of carbon dioxide ceases. The qualification "lower" means that the group in question has no more than four carbon atoms. Compounds of the general formula VIII above may be made by known methods for the preparation of aminoalkyl phenthiazine-10-carboxylates, e.g. by reacting a halide (orester) of a phenthiazinyl-10-carboxylic acid with a compound of the general formula: <FORM:0856529/IV(b)/8> Specification 813,025 is referred to. Reference has been directed by the Comptroller to Specifications 802,725 and 814,512.ALSO:Pharmaceutical compositions comprise at least one compound of the general formula: <FORM:0856529/VI/1> or an acid addition or quaternary ammonium salt thereof (wherein X is a hydrogen or halogen-atom, a lower alkyl or alkoxy group, or a cyano, methylthio, methanesulphonyl, dimethylsulphamoyl or trifluoromethyl group; R is a hydrogen atom or methyl group; and R1 is a lower alkyl group) in association with a significant amount of a pharmaceutical carrier. They may be administered orally or parenterally and may in the form of liquids (including sterile solutions), solids or any combination of these forms. (The qualification "lower" means that the group in question has no more than four carbon atoms). Specified salts are hydrohalides, phosphates, nitrates, sulphates, maleates, fumarates, citrates, tartrates, methanesulphonates and ethandisulphonates, and quaternary salts derived from methyl, ethyl, alkyl and benzyl halides and toluenep-sulphonates. Specification 813,025 is referred to. Reference has been directed by the Comptroller to Specifications 802,725 and 814,512.
