Abstract
The invention comprises racemates and optically active isomers of compounds of the general formula: <FORM:0843070/IV(b)/1> and acid-addition salts thereof, wherein Ar represents a phenyl group or a phenyl group substituted by one or more halogen atoms, alkoxy groups of up to 4 carbon atoms, or methylene dioxy groups, and R represents a hydrogen atom or an alkyl or acyl group of up to 4 carbon atoms; the preparation thereof by benzylating appropriate optical isomers of compounds of the formula: <FORM:0843070/IV(b)/2> or, when R is an alkyl or acyl group, by etherifying or esterifying a corresponding compound wherein R is hydrogen, or an acid-addition salt thereof, and, if desired, converting the products into their acid-addition salts; and also pharmaceutical compositions (see Group VI) containing the products, which are analeptics. 2-Piperidyl-phenyl-methanols of the second general formula above, where R is hydrogen, are prepared by hydrogenation of the corresponding a -hydroxy-benzyl- or benzoyl-pyridines; the erythro forms are separated from racemic mixtures by selective crystallization of their hydrochloride salts, and threo forms are obtained by treating racemic mixtures with SOCl2 and hydrolysing the products. (3,4 - Dimethyloxyphenyl) - (2 - pyridyl) - methanol is prepared by reacting veratricaldehyde and picolinic acid.ALSO:Pharmaceutical compositions which are analeptics comprise racemates and optically active isomers of compounds of the general formula: <FORM:0843070/VI/1> and acid-addition salts thereof, wherein Ar represents a phenyl group unsubstituted or substituted by one or more holagen atoms, alkoxy groups of up to 4 carbon atoms, and methylene dioxy groups and R represents a hydrogen atom or an alkyl or acyl group of up to 4 carbon atoms, with a pharmaceutical diluent. The preparations may be in the form of solutions, suspensions, syrups, elixirs, emulsions, powders, tablets, and capsules. The salt may be the hydrochloride, sulphate, cartrate, citrate or oxalate.
