Abstract

The invention comprises compounds of the formula <FORM:0832490/IV (b)/1> wherein R1 is a phenyl group bearing alkyl or alkoxy substituents of 1-6 carbon atoms, R2 is hydrogen or an alkyl group of 1-6 carbon atoms and R3 is phenyl, esters of the compounds with saturated fatty acids of 2-7 carbon atoms and acid addition salts of these esters, and their preparation by reacting the appropriate N-phenylethyl-piperidone-(4) with an organometallic derivative of an alkyl- or alkoxy-substituted benzene, for example an organolithium derivative, or in the case of the esters, acylating the compound of the formula I by refluxing with a lower fatty anhydride in pyridine. The compounds are also prepared by reacting a styrene derivative R1-C(CH2.R2)= CH2 with an amine R3.CH2CH2NH2 in th presence of aqueous formaldehyde. Products are specified in which R1 and R2 respectively have the values (1) m-tolyl, H; (2) o-tolyl, CH3; (3) m-tolyl, CH3; (4) p-tolyl, CH3; (5) o-ethoxyphenyl, CH3; (6) o-methoxyphenyl, CH3; (7) o-tolyl, C2H5; (8) o-tolyl, C3H7 (n); (9) o-ethylphenyl, H. Acetyl derivatives of these compounds are also referred to. N - Phenethyl - piperidones - (4) optionally bearing alkyl substituents in the 3-position, used as starting materials, are made by cyclizing an amine of the formula R3CH2CH2N.(CH2CH2COO.Alkyl) CH2CHR2COOAlkyl e.g. by treatment with sodium in xylene and treating the resulting 5-carbalkoxy-N-phenethylpiperidone-(4) with aqueous hydrochloric acid to give the desired piperidone or in the case where R2 is hydrogen, the intermediate ketal which is subsequently hydrolysed. Tertiary amines of the formula II are made by treating a primary amine R3.CH2.CH2.NH2 with an acrylic ester which may give the tertiary amine directly or in the case when R2 is an alkyl group, the secondary amine is first formed and is treated with an a -substituted acrylic ester. The Provisional Specification relates generally to compounds wherein R1 is an alkyl, aryl or aralkyl radical.
