Abstract

Solutions of e.g. therapeutic compositions containing components which if mixed in the presence of the residuum of solvent evaporated in freeze drying are gradually inactivated, are successively frozen in layers and the frozen layers are freeze-dried. An insulating layer of solvent only or a solution of an inert material such as lactose, salt or a buffer for H ions may be interposed between the layers. According to an example, an aqueous solution containing riboflavin, nicotinamide, and pantothenyl alcohol is frozen in a vial at - 40 DEG C. To the frozen product there is added an aqueous solution, at a temperature of + 5 DEG C. containing riboflavin, nicotinamide, pyridoxine hydrochloride, thiamine hydrochloride, and Vitamin B12 and the vial is again cooled to - 40 DEG C. to freeze the second solution. The frozen layered cake is freeze-dried using temperatures within the range - 30 DEG C. and + 30 DEG C. and between 50 and 200 microns of mercury pressure.
