Abstract

17-Ketosteroids are produced by bringing into contact a fungus of the genus Gliocladium, or steroid fermentation enzymes derived therefrom, with a 20-oxygenated steroid under aerobic conditions. The process may be carried out by growing the fungus under aerobic conditions, preferably with agitation, in the presence of the 20-oxygenated steroid and of a nutrient medium preferably containing assimilable non-steroidal carbon (e.g. carbohydrate), assimilable nitrogen and phosphorus, and, if desired, other additions such as magnesium sulphate. Preferred fungi are those of the species G. catenulatum, G. roseum, G. deliquescens, and G. luteolum. Examples describe the use of these species in the production of: (1) 4-androstene-3 : 17-dione and its 6b -hydroxy derivative from progesterone, and (2)-(5) from 11-desoxycorticosterone and its acetate or other esters; (6) adrenosterone from 11-ketoprogesterone; (7) 4-androstene-3 : 17-dione from 21-methylprogesterone; (8) 3-hydroxyandrostan - 17 - one from 3b : 17a : 20-allopregnanetriol; (9) 3b - hydroxy - 5 - androsten - 17 - one; (10) etiocholane - 3 : 16 : 17 - trione from pregnane-3 : 6 : 20-trione; (11) 6b -hydroxy - 4 - androstene - 3 : 17 - dione from 6b - hydroxyprogesterone; (12) etiocholane - 3 : 17-dione from pregnane-3 : 20-dione; (13) androstane-3 : 11 : 17-trione from allopregnane-3 : 11 : 20-trione; (14) 4-androstene-3 : 17-dione and its 6b -hydroxy derivative from 17a : 21-dihydroxy - 4 - pregnene - 3 : 20 - dione; (15) adrenosterone from cortisone; and (16) from cortisone acetate; (17) 11a -hydroxy-4-androstene 3 : 17 - dione from 11a : 17a : 21 - trihydroxy - 4 - pregnene - 3 : 20 - dione; (18) 3b -hydroxy - 5 - androsten - 17 - one from 3b - hydroxy - 21 - ethyl - 5 - pregnen - 20 - one. Lists are given of additional starting materials and, in some cases, the products obtainable therefrom, and of other species of Gliocladium.
