719,886. Therapeutic substance from amniotic liquid. THOMAS, A. J. P. J., and THIERY, J. P. July 15, 1952 [July 24, 1951; July 3, 1952], No. 17883/52. Class 81(1) A liquid having valuable biological properties is prepared from amniotic liquid by removing suspended solids, deproteinizing at room temperature, - preferably by foaming, passing over an adsorbent until the liquid no longer gives a precipitate with sulphosalicylic acid and sterilizing at room temperature. As shown, amniotic liquid is centrifuged in a turbo filter comprising a covered tank 1 and a centrifuge bowl with vertical walls 2 of a filter material. The centrifugate containing much foam flows into a receiver 7 wherein the foam 8 is destroyed by gas jets 9 and protein scum is removed through the removable lid 12. The deproteinized liquid flows into a vessel 14 in the bottom of which is a layer of absorbent 15 for polypeptides, e.g. active carbon, and then into a collecting tank 17. Prior to filling into, e.g. ampoules, the liquid is passed through a bacteria-proof filter. Alternatively, the sterile liquid may be dried by lyophilization. Foaming may also be brought about by bubbling through the liquid filtered air, nitrogen, carbon dioxide gas or oxygen. Defoaming may be by means of a defoaming agent. The pH of the liquid is maintained at pH 7.0 by the addition of buffers. Sterilization may be by ultra-violet irradiation. Optionally, prior to the adsorbent treatment, the liquid may be subjected to fractional defreezing.
