Abstract
An antibiotic rhodomycin is produced by growing Streptomyces purpurascens NRRL-2269 under aerobic conditions in a nutrient medium. Rhodomycin is amphoteric; empirical formula C20H29O7N; consists of two components having average molecular weight of 420; contains neither sulphur nor methoxy groups; yields red-coloured solutions in acidic media, purple-coloured solutions in neutral media and blue-coloured solutions in alkaline media; soluble at the isoelectric point (pH8.4-8.6) in alkanols having up to 4 carbon atoms, chloroform, ether, water and acetone; is precipitated from aqueous solutions by picric acid, picrolonic acid and perchloric acid; decomposes when heated with aqueous acidic media; forms salts with mineral salts and with alkalies; absorption band maxima in benzene at 575,542, 535, 504 millimicrons, in acetic anhydridepyroboric acetate mixture at 587, 542 and 505; effective against Staphylococcus aureus in dilutions of 1 in 30,000,000. Salts are also formed with stannous chloride, mercury chloride, chlorauric acid, potassium chromate, chloroplatinic acid, helianthin, alizarine sulphonic acid, anthraquinone sulphonic acid, iodine in potassium iodide, phosphotungstic acid, phosphomolybdic acid, b -naphthalene sulphonic acid, styphnic acid and strong mineral acids, e.g. hydrochloric, phosphoric and sulphuric. The nutrient medium contains a carbon source, e.g. glycerin, glucose or dextrin, a nitrogen source, e.g. glycocoll, alkali metal nitrates, glycine ethyl ester hydrochloride or tyrosine, and mineral sources, e.g. sodium chloride, secondary potassium phosphate, ammonium sulphate, ferrous sulphate, calcium carbonate and Hoagland's solution. Yeast liquor or corn steep liquor may be present. Rhodomycins are obtained from the culture solution or mycelium. The culture solution is concentrated, acetone and basic lead acetate added, and extracted with butanol at pH 8.6. Butanol solution is extracted with 10 per cent acetic acid and the rhodomycins precipitated as picrate. The powdered mycelium is extracted with acidified acetone water or acidified methanol-water, the mycelium centrifuged off, and impurities precipitated from the neutralized centrifugate from which the organic solvent has been removed, by basic lead acetate. The aqueous filtrate is extracted with butanol, and then the butanol extract is extracted with aqueous acetic acid. The antibiotic is precipitated from the acid extract by picric acid.
