Abstract

Hyaluronidase is prepared from finely-divided mammalian testicular tissue by adding water thereto, adjusting the pH of the aqueous mixture to 4.5-4.7 (preferably 4.6), separating the solid material after prolonged stirring (preferably for at least 12 hours), adjusting the pH of the liquid to 7.5-9.2 (preferably 8.0-8.2), separating the resulting precipitate of inactive solid material, adjusting the pH of the liquid to 5.9-6.3, again separating precipitated inactive solid material, adjusting the pH of the liquid to 4.6-7.0 (preferably 4.6-4.8), adding alcohol in such amount that the volume percentage thereof in the liquid is 20-30 (preferably 27-30), separating the further precipitate of inactive solid material, and adding more ethyl alcohol to the liquid to increase the volume percentage thereof to 45-55, thereby producing a precipitate containing hyaluronidase, which is separated from the liquid and may be redissolved in water and the solution diolysed against running water, filtered and evaporated to dryness, the whole process being conducted at a temperature below 5 DEG C. An example describes the application of the process to bull testes, which are prepared for treatment by freezing in dry ice, removing the outer membrane, grinding to powder in the presence of dry ice, allowing the dry ice to evaporate and further grinding the material in a Waring blender.
