Abstract
7 - Hydroxy - and 7 - amino - 1 : 3 : 4-triazaindolizines having fused-on rings in the 5 : 6-positions and of the formula <FORM:0627602/IV (b)/1> wherein R is H or an alkyl, alicyclic, aryl, or heterocyclic group, Y is NH2 or OH, and Z represents the atoms to complete a 5- or 6-membered alicyclic or heterocyclic ring are obtained by heating either a cyclic b -ketoester or a cyclic b -ketonitrile with a 3-amino-1 : 2 : 4-triazole of formula <FORM:0627602/IV (b)/2> wherein R is H or an alkyl or aryl ring. The reactants may be refluxed together in a solvent such as glacial acetic acid, pyridine, or alcoholic alkali. b -Keto esters specified are 1-carbethoxy-2 - ketocyclopentane, 1 - carbethoxy - 2 - ketocyclohexane, 1 - carbethoxy - 2 - phenylcyclohexane - 4 : 6 - dione, 2 - carbethoxy - 3 : 4-diketo - 1 : 5 - diphenylpyrrolidine, 1 - carbethoxy - 2 - Beto - 3 - methyl - 3 : 6 - dimethylmethanocyclohexane, 1 - carbethoxy - 2 - keto-5 - phenylpyrrolidine, 1 - phenyl - 2 - keto - 3-carbethoxypyrrazolidine, 1 - carbethoxy - 6 : 6-dimethyl - 2 : 4 - diketocyclohexane, 2 - carbethoxy - 5 - phenylthiolane - 3 - one, and 2 - (a - furyl) - 1 - carbethoxy - 4 : 6 - diketocyclohexane, b -Ketonitriles specified are 1-cyano - 2 - phenylcyclohexane - 4 : 6 - dione, 1 - cyano - 2 - cyclopentanone, 1 - cyano - 2 - cyclohexanone, and 1-cyano-2 : 2 - di - methylcyclohexane - 4 - 6 - dione. 3 - Amino - 1 : 2 : 4 - triazole or the 5-methyl-, 5-ethyl-, 5-n-propyl, 5-isopropyl -, or 5 - phenyl derivative may be used.
