Abstract

Immunizing substances or antibodies are obtained from the urine of human beings or animals during convalescence from infectious disease. The urine, if desired after filtration and dialysis, may be treated with an albumen precipitant and the antibody recovered from the precipitate, or the antibodies may be extracted from the urine by means of adsorbents, or urine after extraction with ether or similar solvent to remove impurities, may be concentrated or may be mixed with an inert medium and dried. The urine is preferably taken from a patient at such period of convalescence that it contains a minimum of the toxin of the disease. Albumen precipitants specified are lead acetate, phospho-tungstic acid, mercuric chloride, sulpho-salicylic acid, formalin, alcohol and ammonium sulphate. Precipitates made with heavy metal salts are treated with sulphuretted hydrogen to remove the heavy metals and the remaining solution may be further purified by dialysis; precipitates made with substances other than heavy metal salts are dialyzed to remove the precipitants and the remaining solid is extracted with salt solution or weakly alkaline Ringers solution to yield the antibody. Adsorbents specified are aluminium hydroxide kaolin, fuller's earth, silica gel, calcium carbonate and calcium phosphate; these may be added as such or may be precipitated in presence of the urine. The adsorbates may be dried for oral administration, or the antibodies may be extracted therefrom by means of physiological salt solution. For removal of impurities from urine it may be extracted with ether, petroleum ether, benzene, acetic ether, or other water-immiscible solvent and the so treated urine may be concentrated and preferably may be distributed on common salt, milk sugar, aluminium hydroxide or other inert medium and dried in a current of air or in vacuo, preferably over sulphuric acid to absorb any ammonia set free. The antibody preparation may be administered orally or by injection or may be atomized for inhalation.
