Abstract
288,637. Ammon, E. von, and Szombathy, K. April 14, 1927, [Convention date]. Lupanine is extracted from seeds of lupins particularly those of lupinus albus by extracting with a solution of a neutral salt sufficiently concentrated to precipitate the conglutin so as to avoid loss thereof. The, solution is treated, e.g. with sulphuric acid, to precipitate the protein dissolved in the salt solution, filtered, and the filtrate rendered alkaline and extracted with benzol from which the lupanine is recovered. Suitable salt solutions for the extraction contain sodium chloride, sodium sulphate, magnesium sulphate, or of calcium chloride. In an example, ground lupin seeds are immersed in water containing calcium chloride. The mixture is allowed to stand and water is added. The ion concentration is increased by the addition of acid, e.g. hydrochloric acid. After several hours the alkaloids are dissolved. The residual seeds containing 25 per cent of liquid and some alkaloid still associated with them are heated with fresh salt solution, after which they are placed in a filter press and finally washed with water at 78‹-82‹ C. The cakes are dried and may be then ground. Sulphuric acid is added to the liquid containing the bulk of the alkaloid to effect the precipitation of the dissolved protein, and the lupanine is extracted with benzol from the solution, filtered and made alkaline, and recovered by crystallization. The lupanine when converted into the hydrochloride can be separated into the d and l forms as the d-lupanine hydrochloric can be dissolved in chloroform while the l-salt remains undissolved.
