Abstract
The invention relates to the delivery of food colorings, flavors, beverages or medicaments in compositions encapsulated in one-piece soft gel capsules. The capsules shell may rupture in contact with warm drink/water over temperature of 26 degrees centigrade and the contents are released immediately or prolonged to achieve desired effects. The capsules may serve to slowly release their contents. The invention offers better consumer convenience and accuracy of the dosage for beverages, flavors or colorings. The carrier material may be polyethylene glycol 400, glycerol esters of fatty acids or gum acacia and the delivered ingredient may be caffeine, tea, coffee, cardamom, capsicum, cinnamon or medicaments, such as analgesics, NSAID, cough medicines and cardiovascular pharmaceuticals.

Moonga, BritHealth Ltd, Business & Technology Centre, Bessemer Drive, Stevenage, Herts., SGl 2DX England Patent Application No. GB 9919323. 7 Filing Date 17"'August 1999 ABSTRACT: The invention relates to the one-piece soft gel capsules and compositions of delivery systems for delivery of medicaments and nutrition substances for absorption in the body through gastrointestinal tract for prolonged action. The other aspect of the invention relates to the delivery of food colorings, flavors and beverages in compositions encapsulated in one-piece soft gel capsules, which can be delivered directly to prepare drinks and foods. The capsules shell ruptures in contact with warm drink/water over temperature of 26 degree centigrade and the contents are released immediately or prolonged to achieve desired effects. The invention offers better consumer convenience and accuracy of the dosage for beverages, flavors or colorings The invention also offers methods for delivery of flavors of over prolonged periods. The invention offers opportunity for pharmaceutical, beverages, foods and flavor industry to formulate new products for consumer's convenience and accurate delivery of the active substance.
BACKGROUND TO THE INVENTION Generally oral administered drug dosage forms include elixirs or drugs in liquid, tablet or hard gelatin capsules containing unit dosage of selected drug requiring dosage of two or three times per day. The slow release enables the drug to be released over an extended period while maintaining sufficient bioavailability. The slow release dosage forms have been used for hard gelatin capsules having enteric-coated pellets therein or compressed into tablets and coated from the outside.
In case of flavors no patent is published to the knowledge of the inventor regarding prolonged release in foods. A Canadian patent No. 888683 relates to Seamless Elastic Gelatin capsule (SEG) dosage form in which the fill material when in contact with water or gastric juice forms micro porous spongy substance, which diffuses the active.
Although the sustained release drug form disclosed in the patent relates to a possible system, there is need and further interest in modified release systems those can be encapsulated in one-piece soft gel capsules. These formulations should deliver the active substance under variable conditions of physiology or environment A US patent dated January 1989 Number 4,800, 083 by Food S etal used fill material of a masticator gum/elastomer having medicament dispersed in the matrix and the active substance absorbed by the body. The system is highly insoluble and mechanism for release was suggested as peristaltic movement in the gastrointestinal tract.
Claims (8)
- CLAIMS The inventor claims ; 1. The methods and compositions of delivery systems for beverages, food colorings and flavors. The active substances are encapsulated in one-piece soft gel capsules and the contents are released in contact with warm water/aq medium of the food or drink. The active substances examined included food colorings, flavors and beverages approved under EEC directive 98.
- 2. The delivery of flavors over extended period by incorporation of one of the followings ; gum acacia, guar gum, chitosan, Gellucire 53/10 (from Gattefose), Methyl cellulose mixed with Gellucire 33/01 and Polyethylene Glycol 400
- 3. The method and composition of delivery systems of drug substances to give prolonged desired effects. The composition included Chitosan or gum acacia or guar gum or Gellucire 53/10 or Glyceryl behenate (from Gattefosse. The active substances examined included Analgesics, NSAID, Cough medicines, and Cardiovascular drugs and some examples include Ibuprofen, Nifedipine, Naproxen, Ketoprofen, theophylene, Cimetidine, Indomethacin and vitamin C etc.The other systems examined included Polyethylene Glycols-Drug substance conjugates suspended in Gellucire 33/01.The viscosity of the formulation was adjusted using triethyl citrate or castor oil or glycerin for encapsulation in soft gel one-piece capsules. The capsules were coated with pH sensitive polymethacrylic/acrylic co-polymers in case of insulin and calcitonin capsules. Pectinate was another material examined for coating of such capsules containing peptide drug substances. The capsules showed delivery of peptides and provided protection in the gut against biological and chemical degradation.
- 4. The use of Eudragit E polymer, Gellucire 33/01 and Polyethylene Glycol 400 for taste and odor masking of drug substances.
- 5. The composition of Tea capsules :INGREDIENTS PER CAPSULE Polyethylene Glycol 400 25mg Gellucire 33/01 20mg Caffeine 20mg Cardamom extract Img Tea colour/flavour, 0.5mg Cinnamon extract 0.5mg Gelatin 54mg Glycerin 25mg Water 8mg The flavor and extracts can be varied to prepare any herbal or other nutritional beverage in capsule form which will dissolve immediately in contact with warm aqueous medium.

The composition of a Coffee Capsules<Desc/Clms Page number 3>INGREDIENTS PER CAPSULE Coffee powder 100mg Polyethylene Glycol 100mg Gellucire 33/01 50mg Gum acacia 5mg Gelatin 100mg Glycerin 50mg Water 15mg The concentration or dosage of the coffee can be varied in proportion with other ingredients for encapsulation in soft gel capsules. The capsules dissolved immediately in contact with warm water and releasing accurate dosage of the coffee and retained freshness/flavor.
- 7. The composition of a food flavorINGREDIENTS PER CAPSULE Cardamom Extract 2mg Polyethylene Glycol 40mg Gellucire 33/01 35mg Gum Acacia 25mg Gelatin 30mg Glycerin 20mg Water 4mgThe cardamom extract can be replaced with any other flavor approved by EEC directive 98.
- 8. The composition of food coloring capsulesINGREDIENTS PER CAPSULE Oleoresin Capsicum 20mg Polyethylene Glycol 40mg Gellucire 33/01 30mg Gelatin 24mg Glycerin 14mg Water 4mg4'Et..... vC. IRE. i. Q. R. c : J' : ;' Tt- M.. o.. t-k" < U<Desc/Clms Page number 4>The oleoresin capsicum can be replaced with any other coloring approved by EEC directive 98. The dosage can also be varied in proportion with other excepients.CONCLUSION AND DISCUSSION The invention offers opportunity to pharmaceutical industry to formulate slow release formulations in a consumers convenience dosage form in one-piece soft gel capsules, which are tamper evident. The invention opens new opportunities for delivery of peptide drugs.The other aspects of the invention offers opportunities to food, beverages and flavor industries to develop new products which gives consumers convenience and accuracy of the dosage for use in delivery of colorings, flavors and preparation of beverages.
