1504412 Triazinobenzodiazepine triones; their preparation and pharmaceutical applications E R SQUIBB & SONS Inc 16 May 1975 [9 Aug 1974] 20912/75 Heading C2C The invention comprises compounds of general Formula (I) wherein R 1 is hydrogen, C 1-4 alkyl, phenyl or phenylmethyl; R 2 is hydrogen or C 1-4 alkyl; R 3 is C 1-4 alkyl or aralkyl and R 4 is hydrogen, halogen, NO 2 , CN, CF 3 , C 1-4 alkyl, C 1-4 alkoxy or C 1-4 alkylthio which may be prepared by reacting a compound of Formula (II) wherein R 5 is halogen, sulphhydryl, alkoxy, alkylthio or phenylalkylthio and R 6 is alkyl, phenyl or phenylmethyl with a compound of formula IIa to obtain a compound of Formula (III) reacting said compound of Formula III with a base (e.g. thallousethoxide) to obtain a salt of Formula V reacting this salt with a compound R 3 -X where X is Cl, Br or I, alkylsulphonate or arylsulphonate and if desired reducing the product to obtain a compound of Formula I in which R 6 is H. 11 - Chloro - 2,8 - dihydro - 8 - methyl - astriazino - [4,3 - d] - [1,4] - benzodiazepine - 3,4,7 (6H)-trione is prepared by reaction of 5,7- dichloro - 1 - methyl - 1,4 - benzodiazepinone - 2 with 4-morpholineglyoxylic acid hydrazine and is reacted with thallous ethoxide to form the thallous salt. 11 - Chloro - 2,8 - dihydro - 8 - phenylmethylas - triazino - [4,3 - d] - [1,4] - benzodiazepine- 3,4,7(6H) - trione is prepared by reaction of 5,7 - dichloro - 1 - benzyl - 1,4 - benzodiazepinone - 2 with 4 - morpholinoglyoxylic acid hydrazide and is reacted with thallous ethoxide to form the thallous salt. Pharmaceutical compositions having antiinflammatory activity for oral or injection application comprise a compound of Formula I.
