Abstract

1492959 Plasimmogen activator preparations PIERRE FABRE SA 3 Dec 1974 [3 Dec 1973] 52216/74 Heading A5B [Also in Division C3] A plasimmogen activator useful in treating arterial and venous thrombosis is prepared by (a) suspending a powdered acetone extract of animal organs in an aqueous alkali metal or ammonium salt solution having an ionic strength in the range 0.01- 0.1 and a pH of 5 to 8, (b) taking up the precipitate so obtained in an aqueous alkali metal or ammonium salt solution having an ionic strength of 0.6 to 1 and a pH of 3 to 5, (c) decanting the solution so obtained and adding a salt or organic solvent at pH 3 to 5 so as to form a preciptate, (d) taking up the precipitate in water or alkali metal or ammonium salt solution and adding a salt at pH 7 or slightly above so as to form a further precipitate and (e) dissolving the precipitate in water and dialysing the solution up to a resistivity level of 1000 ohm. cm at 10‹C. The animal organs may be sow ovaries. The reagents used may be in step (a) potassium chloride or acetate, in step (b) potassium chloride, acetate or sulphate, in step (c) ammonium sulphate, potassium chloride or acetone and in step (d) sodium chloride, ammonium sulphate or potassium sulphate. The product may be purified by salt precipitation, chromatography in a mixed dextramagarose and polyacrylamide gel or ion-exchange chromatography.
