Abstract

1487110 Determination of cholesterol ONO PHARMACEUTICAL CO Ltd 22 May 1975 [22 May 1974] 22089/75 Heading G1B Bound and free cholesterol in an animal serum sample are determined by a method in which a sample of the serum is treated and then analysed for free cholesterol and another sample is analysed for free cholesterol after treatment which is similar but which includes treatment with cholesterol esterase to convert bound cholesterol to free cholesterol, the cholesterol esterase being obtained from Pseudomonas fluorescens. In analysing for free cholesterol, the sample is treated with 1% digitonin in 50% alcohol and centrifuged, then to the precipitate is added glacial acetic acid and ferric chloride in sulphonic acid solution, and the extinction coefficient at 560 mn is determined.
