Abstract

1485340 Determination of thyroid content utilizing pepsin digestion CENTER FOR LABORATORY MEDICINE 25 July 1975 31257/75 Heading G1B A process for the determination of thyroid hormone content of a blood serum sample comprises mixing with a sample of blood serum pepsin at a pH of about 1À0, incubating the sample for a period of time and at a temperature sufficient to allow the pepsin to digest the thyroid hormone binding protein in the serum, adjusting the mixture to a pH at which the pepsin is inactive and assaying the mixture for the released thyroid hormone. The pepsin may be in the form of an aqueous solution containing 6À0N.HCl or alternatively glycine hydrochloride, glutamic acid hydrochloride or aspartic acid hydrochloride, or pepsin and an acid may be included in a tablet comprising pepsin, glutamic acid hydrochloride and lactose. A process utilizing the invention in radioimmuno assay of I-125-thyroxine is described.
