Abstract
1481809 Cephem derivatives PFIZER Inc 14 Oct 1974 [17 Oct 1973] 22872/76 Divided out of 1481807 Heading C2C The invention comprises compounds of the formula and salts thereof, wherein T is a tetrazolyl group which is either wherein R is H or 2,2,2-trichloro- or -bromoethoxycarbonyl, benzyloxycarbonyl or wherein R 3 , R 4 and R 5 are H, Cl, Br, F, CH 3 , CH 3 0 or phenyl; A is H, CH 3 COO, 1-methyl-5 tetrazolylthio or 2-methyl-1,3,4.thiadiazolyl-5- thio; R 1 is H, (C 3 -C 6 )alkanoyloxymethyl, 1-[(C 4 -C 7 )alkanoyloxy]ethyl, methoxymethyl or phthalidyl; and R 11 is the same as R 1 or is selected from groups of the formulµ and wherein R 6 is (C 1 -C 3 )alkyl or phenyl, R 7 is OH, CH 3 0, (C 2 -C 4 ) alkanoyloxy or benzyloxy, R 8 is H, OH, F, Cl, Br, I, CH 3 , CH 3 0, (C 2 -C 4 ). alkanoyloxy, phenyl or benzyloxy, R 9 and R 10 are each H or CH 3 and X is O or S. They may be obtained by reacting a compound of Formula I above except that T is replaced by -CONHR 2 wherein R 2 is selected from groups of Formulµ III and IV above, with a chlorinating agent to produce an imino chloride followed by reaction with an azide to form a tetrazolyl group; optionally replacing the R 2 group by treatment with a mixture of CF 3 COOH and anisole. Compounds in which R 1 or R 11 is other than H may be obtained by alkylating a compound in which R 1 or R 11 is H with a compound of formula to form a compound wherein R 1 is other than H. Compounds in which A is other than hydrogen may be obtained by brominating compounds in which A is H with N-bromosuccinimide and reacting the resulting 3-bromomethyl compound without isolation with an acetate salt or an appropriate mercaptan. Compounds in which R is H are obtained from (a) compounds in which R is the 2,2,2-trihaloethoxycarbonyl group by treatment with Zn dust and acetic acid, (b) compounds in which R is the triphenylmethyl group by treatment with HCOOH, and (c) compounds in which R is benzyloxycarbonyl group by treatment with a mixture of CF 3 COOH/anisole and CF 3 S0 3 H. The products are intermediates useful in the preparation of antibacterial agents.
