Abstract
1478460 Glucopeptides E FISHER J L J SZABO and N SZAPANOS 19 Aug 1974 [29 Aug 1973] 40612/73 Heading C3H Hypolipaemic glucopeptides are prepared by freeing bovine duodenum material in small particles from fat, hydrolysing the animal tissue in an aqueous medium which contains an alkali or an enzyme, e.g. papain, separating undispersed particles by filtration or centrifuging, precipitating crude glucopeptide from the residual liquid by addition of alcohol and then purifying the crude glucopeptide. The alkaline medium may be sodium hydroxide of IN concentration and hydrolysis is continued for 4 days at room temperature, followed by neutralization to a pH from 6À5 to 7À5. Hydrochloric acid may be used for neutralization to a pH of from 7 to 7À5 or acetic acid may be used for neutralization to a pH of from 6À5 to 7. The crude precipitated glucopeptide may be dissolved in sodium chloride solution, the solution clarified by centrifuging, a precipitant then being added to the supernatant, removing the resultant precipitate, dissolving the precipitate in sodium chloride solution, clarifying the solution by centrifuging, adding alcohol to the supernatant and separating the final precipitate by filtering or centrifuging. The precipitant may be Cetrimide, trichloracetic acid or cetyl pyridinium chloride. Preferably the crude glucopeptide is purified by drying, mixing and pulverizing with cellulose powder or diatomaceous earth, charging a chromatographic column with the mixture, passing a solvent through the column, eluting the glucopeptide with a phosphate buffer at pH 7À8 and precipitating the glucopeptide with two volumes of alcohol, and filtering the purified glucopeptide. The purified glucopeptide may be dried, ground and screened.
