Abstract

1458147 Detection of microbial pathogens in fluid J K & SUSIE L WADLEY RESEARCH INSTITUTE & BLOOD BANK 7 Feb 1974 [9 Jan 1974 12 Feb 1973] 05685/74 Heading GIB [Also in Division C6] Microbial pathogens in a fluid are detected by depositing acid fluid upon a liquid of greater density and centrifuging, pathogens thereby passing into the liquid. The liquid may be an aqueous solution of non-ionic sugar, e.g. sucrose, or a macromolecular solute, e.g. a copolymer of sucrose and epichlorohydrin, or dextran, of molecular weight 10,000-2,000,000. The fluid may be a body fluid e.g. blood, bone marrow, spinal fluid, pleural fluid, or urine. The liquid may be separated, mixed, and plated out on a nutrient medium for microscopic analysis. In a modification, a gelling agent is added to the liquid in a centrifuge tube, which is then cooled to solidify the liquid, then the fluid is added and the tube warmed to liquefy the gel for centrifuging. The gelling agent may be gelatine. A device for use in the method comprises a tube closed by means allowing passage of a hypodermic needle and containing a sterile aqueous solution for reception of the micro organisms, the aqueous solution comprising less then 50 weight per cent of a gelling agent, which may be an epichlorhydrinsucrose polymer or a dextran.
