Abstract

1454529 Thiophene derivatives SANDOZ Ltd 27 Nov 1973 [1 Dec 1972 14 Dec 1972] 54968/73 Heading C2C A compound of Formula III is made by reaction of a compound of Formula I with a compound of Formula II where A 1 is a radical whose activity does not compete with that of the rest of the molecule I, and A 2 is a radical whose activity does not compete with the rest of II but provides the vicinal methylene group with sufficient acidity to react with the =NH group of I. A 1 is for example C 1 -C 8 alkyl, or phenyl with substituents selected from F, Cl, Br, C 1 -C 3 alkyl, CN and NO 2 . A 2 is for example phenyl with at least one substitutent selected from NO 2 , CN and alkylsulphonyl in the ortho or para position, CN, C 1 -C 4 alkoxy, phenoxy or substituted or unsubstituted carbamoyl, thiocarbamoyl, alkoxycarbonyl aryloxycarbonyl, alkylcarbonyl or benzoyl. III in which A 2 is carbamoyl may be reacted with phosphorus pentasulphate to form III in which A 2 is thiocarbamoyl. A 2 in III may also be converted from alkoxycarbonyl to -COOH by saponification, or to -H by saponification followed by decarboxylation. In examples, when A 1 is CH 3 , A 2 is CN, CH 3 OCO, C 2 H 5 OCO, NH 2 CO, NO 2 Ph, NH 2 CS, HOOC and H; when A 1 is Ph, A 2 is CN and CH 3 OCO.
