1447218 Phenoxy alkanols and esters thereof MERCK PATENT GmbH 26 March 1975 [2 April 1974] 12745/75 Heading C2C Novel compounds I: wherein R<SP>1</SP> is H or CH 3 ; R<SP>2</SP> is CH 3 or C 6 H 5 ; R<SP>3</SP> is 4-halo-phenoxymethyl, 1,2,3,4-tetrahydro-1- naphthyl, 1-pyrryl, piperidino, isoindolino, 1,2,3,4-tetrahydroquinolino, 1,2,3,4-tetrahydro- 1 - R<SP>5</SP> - 4 - quinolyl, 4 - chromanyl, 4 - thiochromanyl, or, if R<SP>1</SP> and R<SP>2</SP> are not both CH 3 , R<SP>3</SP> may also be hal, phenyl, 4-hal-phenyl or 4- hal-phenoxy; R<SP>4</SP> is H or alkanoyl with 2 to 4 carbon atoms; R<SP>5</SP> is H or CH 3 and hal is F, Cl or Br; and their physiologically acceptable acid addition salts, are prepared by (i) reacting a phenol II: (which may be partly or wholly in the form of a phenolate) with a compound of formula X-CR<SP>1</SP>R<SP>2</SP>-CH 2 OR<SP>4</SP> in which X is Cl, Br, I, OH or an esterified OH group provided that when X is OH, R<SP>4</SP> is alkanoyl with 2 to 4 carbon atoms, to form a compound of Formula I, or (ii) reducing a compound of Formula IV: wherein Y is a free or functionally modified COOH group, a free or functionally modified CHO group, or a CH 2 OCH 2 C 6 H 5 group, to form a compound of Formula I, or (iii) treating a compound of Formula IV above wherein Y is a CH 2 hal group or an esterified or etherified CH 2 OH group, with a solvolysing agent, to form a compound of Formula I, or (iv) diazotizing a compound of Formula IV above wherein Y is an aminomethyl group, and reacting the resulting diazonium compound with a compound of formula R<SP>4</SP>-OH wherein R<SP>4</SP> has the abovestated meaning, to form a compound of Formula I, and, if desired, converting a compound I into another compound I by saponifying an alkanoyloxy group therein and/or esterifying a hydroxyl group therein. A resulting compound I may be reacted with an acid to form a physiologically acceptable acid addition salt. Pharmaceutical compositions for oral, parenteral or topical administration comprise a compound of the invention together with a pharmaceutical carrier. The compositions are useful for lowering cholesterol, triglyceride and uric acid levels and for inducing liver enzymes.
