Abstract
1438130 Prostanoic acid derivatives ROUSSEL UCLAF 27 Dec 1973 [27 Dec 1972] 59819/73 Heading C2C Novel compounds I in which R is H or C 1-4 alkyl, m is 3, 4 or 5, n is 2, 3 or 4, R<SP>1</SP> is a saturated or unsaturated, acyclic C 1-4 hydrocarbon radical or a C 3-6 cycloalkyl or C 3-6 cycloalkenyl radical, A is C 1-4 alkoxy or keto, and both of the symbols = in the 5- membered ring represent single bonds when A is keto or one of the symbols represents a single bond and the other symbol represents a double bond when A is C 1-4 alkoxy, and base salts of those compounds I in which R is H, are prepared by treating a compound II in which each of alc and alc 1 is C 1-4 alkyl, with an acid to obtain a compound III which is treated with a C 1-4 diazoalkane to obtain a compound IV in which A is C 1-4 alkoxy, which is saponified with an alkaline base to obtain, after acidification, a compound V which is heated to effect decarboxylation and, if desired, treated with a C 1-4 diazoalkane to obtain a mixture of compounds VI and VI<SP>1</SP> if desired, separating the constituents compounds of the mixture, treating the mixture or a constituent compound thereof with an oxidizing agent to obtain a mixture of compounds VII and VII<SP>1</SP> or a constituent compound thereof, if desired, separating the constituent compounds of the mixture, treating the mixture or a constituent compound thereof with a compound of Formula R<SP>1</SP>MgX in which X is halogen, to obtain a mixture of compounds IX and IX<SP>1</SP> or a constituent compound thereof, if desired, separating the constituent compounds thereof and, if desired, reacting the mixture or a constituent compound thereof with an acid to obtain a mixture of compounds X and X<SP>1</SP> or a constituent compound thereof, the said mixture being separated, if desired, into its constituents, and, if desired, converting a resulting compound I in which R is H into a base salt thereof. Pharmaceutical compositions having hypotensive, antibionchoconstrictive and a contracting effect on the smooth muscle, for oral, parenteral, rectal or topical administration, comprise a compound of the invention together with a pharmaceutical carrier.
