Abstract

1430238 Preparation of esters of N-blocked amino acids GLAXO LABORATORIES Ltd 4 July 1973 [10 July 1972] 32213/72 Heading C2C A process for the preparation of an ester of an N-blocked amino acid, for example, a penicillin or cephalosporin, comprises reacting the N- blocked amino acid, or a salt thereof, with an amine of formula (where R is an organic group) and a nitrosating agent to yield an N-blocked amino acid ester in which the ester group has the formula A number of penicillins and cephalosporins are prepared by the process and the following compounds are stated to be novel:- 2-furylmethyl - 3 - acetoxymethyl - 7# - (2 - thienylacetamido)ceph - 3 - em - 4 - carboxylate; cyclohexyl - 3 - acetoxymethyl - 7# - (2 - thienylacetamido)ceph - 3 - em - 4 - carboxylate; benzyl - 3 - acetoxy - methyl - 7# - (2 - thienylacetamido)ceph - 3 - em - 4 - carboxylate; 9 - fluorenyl - 3 - methyl - 7# - phenoxyacetamidoceph - 3 - em - 4 - carboxylate; and triphenylmethyl - 3 - methyl -7# - phenoxyacetamidoceph - 3 - em - 4 - carboxylate. The compound diphenylmethyl D-(-)-N-(2,2,2- trichloroethoxycarbonyl) - 2 - phenylglycinate is also prepared.
