Abstract

1424427 Calcium determination DOW CHEMICAL CO 13 July 1973 [11 April 1973] 33490/73 Heading G1B A composition for the determination of calcium in biological fluids comprises a solution of orthocresolphthalein complexone, in an aqueous alkaline amine base buffer, wherein said base is an amino lower alkanol of three or four carbon atoms having a pK in water of from 9À3 to 10, the composition having a pH of 10À1 to 10À7. The preferred base is 2-ethylaminoethanol. The composition may further comprise sodium or potassuim cyanide, 8-hydroxy quinaline and a water-soluble surfactant. The calcium content of a biological fluid such as blood serum is determined by mixing the fluid with the composition and incubating for a suitable time e.g. 2-10 mins, the resulting colour change due to the presence of calcium being measured, for example photometrically. The composition may be formulated in kit form, and may comprise two separate aqueous reagents, one being an acidic solution of the complexone, and the other solution being an alkaline solution of the amino lower alcohol.
