Abstract

1411421 Urokinase-heparin complex CHOAY SA 22 Sept 1972 [24 Sept 1971 30 June 1972 (2)] 44003/72 Heading C3H [Also in Division A5] A complex of urokinase and heparin is prepared by admixing a solution of urokinase and a solution of heparin, adjusting the pH of the mixed solutions, e.g. with phosphoric acid, to from 1 to 5, preferably about 4.2, for formation and precipitation of the complex and, if desired, freeze-drying the complex. The complex may have typical relative activities of 30,000-100,000 CTA units of -urokinase per 100 IU of heparin. A solution for perfusion comprises the complex dissolved in a pharmaceutically acceptable, isotonic solution, e.g. 5% glucose serum, which is compatible with urokinase, does not form a precipitate with heparin and is free of mineral salts. The solutions may have urokinase concentrations of the order 300,000-900,000 CTA units/250 ml. Methods for obtaining a pyrogenic urokinase as starting material for the complex formation are described in detail. They may involve extracting crude urokinase from human urine by absorption on diatomaceous earth and elution of urokinase therefrom, elimination of viral contamination as by heating to 60‹C. under slightly acid conditions and cooling and separating off the solid residue, and two steps of fractionation using diethylaminoethyl cellulose employing in the first step suspensions of pH 4-6 and conductivity 15,000-25,000 micromhos/cm. with 15-40 g. of resin per 14 million CTA units of urokinase, and in the second step suspensions of pH 6-7 and conductivity 15,000-18,000 micromhos/cm. with 30-60 g. of resin per 10 million CTA units of the enzyme. The resulting urokinase-containing medium is diluted with quantities of water sufficient to lower the conductivity by about 1000 microinhos/cm. at each successive dilution until a suspension having a conductivity of about 9,000 micromhos/cm. is obtained.
