1375986 Macrolide antibiotic compositions JULIUS SCHMID INC 23 Nov 1971 54462/71 Heading ASB A macrolide antibiotic dosage form consists of a pharmaceutical capsule containing a multiplicity of beadlets (which term includes beads, granules, and pilules) which are composed of: (i) a polyenic macrolide and (ii) an absorbent material capable of binding the macrolide under acid conditions and releasing it under substantially neutral conditions, the weight ratio of (i) to (ii) being from 1:2 to 1:8. The macrolide may be a hexaene or heptaene macrolide e.g. candicidin, fungimycin, hamycin or mediocidin and the absorbent material may be a salt or base of a di- or tri-valent metal e.g. a carbonate, phosphate, sulphate, silicate or hydroxide of Ca, Al, Mg, Bi or Fe, a cellulose derivative, gum or ion exchange resin. The beadlets may be prepared by known methods and may be given an enteric coating, e.g. of a fatty or waxy material.
