1374918 Triiodinated amide substituted benzenes LABORATORIES ANDRE GUERBET 7 March 1972 [7 April 1971] 10515/72 Heading C2C [Also in Division A5] The invention comprises compounds of the formula in which R 1 is an alkylene radical, R 2 and R 3 are alkyl radicals, X is OH or NH 2 , Y is OH or -NHR 4 , R 4 being an alkyl group, and ammonium, alkali metal, alkaline earth metal or organic base salts thereof. The compounds are prepared by reacting an amide of formula with a halogenated compound of formula wherein Z is halogen optionally in a solvent. It is preferred that alkyl and alkylene groups have 1-6 carbon atoms and the preferred salts of compounds of Formula I are the methylglucamine salts. Exemplified compounds are 3 - (N - acetylcarboxymethylamino) - 2,4, 6- triiodo - 5 - (N - methylcarbamyl) - benzoic acid, 3 - (N - acetylcarbamyl methylamino)- 2,4,6 - triiodo - 5 - (N - methylcarbamyl)- benzoic acid and 2,4,6-triiodo-1,5-di-(N- methylcarbamyl) - 3 - (N - acetylcarboxymethylamino)-benzene. 2,4,6 - Truodo - 3,5 - di - (N - methylcarbamyl)-acetanilide is prepared from the corresponding aniline by reaction with acetyl chloride and is subsequently reacted with bromoacetic acid to form the methyl ester of 2,4,6-triiodo-1,5- di - (N - methylcarbamyl) - 3 - (N - acetylcarboxymethylamino)benzene. The compounds of the invention are of value as radiation opacifiers in Urography, Angiography and Myelography. and may be applied as aqueous solutions, optionally together with preservatives, after adjustment to pH 7, by intravascular injection or by the oral or retrograde routes.
