Abstract

1363382 1,1-Diphenyl-1-(2-imidazolyl)carbinols GIST-BROCADES NV 2 Feb 1973 [4 Feb 1972 5 Sept 1972] 5418/72 and 41183/72 Heading C2C [Also in Division C3] The invention comprises compounds of formula and their acid addition salts wherein one of R 1 ... R 10 is halogen, CF 3 or Me 3 C and the others may be the same or H; R 11 and R 12 are each H, C 1-6 alkyl or Ph (possibly substituted by halogen or CF 3 ); and R 13 is H, C 1-6 alkyl (=R), ROCH 2 , Ph*-C 1-6 -alkyl, C 2-6 alkenyl, Ph*-C 1-6 - alkoxy CH 2 (Ph* being Ph optionally substituted by halogen, CF 3 or R), or PhSO 2 (possibly R-substituted). In examples, these compounds are prepared by (1) splitting-off R 13 (a) hydrolytically, when this is tosyl, CH 2 OMe or of (b) oxidatively, when this is alkenyl; (2) N- substitution of the compound where R 13 = H; and (3) reaction of (a) a corresponding imidazolyl phenyl ketone or benzophenone with a Grignard or organo-Li reagent derived from (respectively) a benzene or imidazole compound, or (b) an ester of (e.g.) 1 - methoxymethylimidazole - 2 - carboxylic acid with such reagent derived from a benzene compound. Intermediates otherwise prepared are imidazoles substituted thus: 1-allyl-4(or 5)-p-chlorophenyl; 1 - p - chlorobenzyl; 1 - benzyloxymethyl; and 1 - methoxmethyl - 4,5 - dimethyl (or diisobutyl). Therapeutic compositions having anorexiant, analgesic, antiphlogistic and antipyretic activity, comprise compounds of the above formula, and may be administered orally or parenterally.
