Abstract

1351547 Enzymatic determination of ethanol WORTHINGTON BIOCHEMICAL CORP 7 March 1972 [22 March 1971] 10589/72 Heading G1B In a method for the enzymatic determination of the ethanol content of an ethanol-containing material, a sample of ethanol-containing material is commingled with an aqueous solution of alcohol dehydrogenase that contains 2-piodophenyl-3-p-nitrophenyl-5-phenyl tetrazolium chloride ("INT") in an amount that in the reaction system is at least sufficient to be quantitatively responsive to the ethanol content of the sample as regards conversion to formazan, nictoinamide-adenine-nucliotide, diaphorase and a buffer which in said solution is substantially inert and which maintains the pH of the solution at a value from 7À5 to 9À5 under testing conditions, The solution may also contain albumin or gelatine as stabilizer and glutathione as antioxidant. A test composition may comprise a lyophilized mixture of the components other than INT, a solution of INT being separately lyophilized. The process is applicable to wine, vinegar or biological fluids e.g. serum.
