1347036 Biphenylene derivatives MERCK & CO Inc 6 Sept 1971 [15 Sept 1970] 41500/71 Heading C2C Novel compounds of Formula I where R is a hydrogen atom or an alkyl, alkoxy, alkenyl, alkylamino or haloalkyl radical; each of R 1 and R 2 , which may be the same as or different from the others, is a hydrogen or halogen atom or a nitro, amino, alkylamino, dialkylamino, alkenyloxy, alkenyl, acylamino, cyano, carboxy, alkoxy carbonyl, carbamoyl, methylenedioxy, trihaloalkyl, alkoxy, alkyl, mercapto, alkylthio, alkylsulfinyl, alkylsulfonyl, sulfonamido, sulfinamido, aryl, aralkyl, aryloxy, aralkoxy, hydroxy, hydroxyalkyl, ahloalkyl, mercaptoalkyl, alkylmercaptoalkyl, aminoalkyl, alkylaminoalkyl, dialkylaminoalkyl, alkoxyalkyl, acyl or acyloxy radical; and Y is a hydroxy, amino, alkylamino, dialkylamino, cycloalkylamino, N-heterocyclo, alkoxy or aralkoxy radical, or a radical of formula-OMe where Me is a non-toxic cation are prepared either by hydrolysis of a novel nitrile of Formula II where R, R 1 and R 2 are as above except that R 1 and R 2 are not cyano, acylamino, carboxy or haloalkyl, prepared by reacting NaCN with the halide Br for CN in II above; or by reacting a compound of Formula III with a secondary amine and sulphur and hydrolysing the reaction product in situ and in both cases optionally converting a group R 2 or R 1 or Y into another by standard methods or forming a salt. Intermediates of Formula IV where R 1 and R 2 are each H, alkyl aryl, aralkyl, halogen or CF 3 are prepared by halogenation of the corresponding alcohols which are obtained by reduction of the acetylbiphenylenes. Pharmaceutical compositions in conventional forms for topical, oral, rectal or parenteral administration and having anti-inflammatory, analgesic and antipyretic activity comprise an above novel compound of Formula I and a carrier or diluent.
