1324060 Nitrofuryl-triazolo(4,3-6)pyridazines BOEHRINGER MANNHEIM GmbH 13 March 1972 [19 March 1971 11 Dec 1971(2)] 11607/72 Heading C2C Novel compounds of general formula (I) in which X is oxygen or sulphur, n is 0 or 1, Hal is an s-triazolo-(4,3-6)pyridazine system and R 1 and R 2 which may be the same or different are a C 5-7 cycloalkyl group or saturated or unsaturated hydrocarbon radicals (which may be substituted by hydroxyl, mercapto, alkoxy, alkylthio, morpholino, furyl aryl or optionally alkylated or acylated amino groups) or NR 1 R 2 represent a piperidine, piperazine, pyrrolidine or morpholine ring (which rings may be substituted by a hydroxyl, carboxyl, alkyl or hydroxyalkyl radical) or one of R 1 and R 2 may represent hydrogen, are prepared by (1) reaction of a compound of general formula II with a compound Y-NR 4 R 5 in which R 4 and R 5 are hydrogen or R 1 and R 2 and Y is hydrogen or formyl and when R 4 and/or R 5 is hydrogen the amino group may be subsequently substituted by one or two of the radicals R 1 and R 2 , (2) nitration of a compound of general formula (III) (3) cyclization of a compound of general formula (IV) in which R 3 is hydrogen, hydroxyl, or amino, (4) reaction of a compound of general formula (V) or (VI) with an aldehyde of general formula VII 3 - Dimethylamino - 6 - (2 - furyl) - s - triazolo- [4,3-6]pyridazine, a novel intermediate for use in method (2), is prepared by cyclization of a tri-carbethoxy derivative of 3-(2-furyl)-6-hydrazino pyridazine to give the 3-hydroxy compound which is reacted with POCl, to give the corresponding chloro compound which is reacted with aqueous dimethylamine. Compounds of general formula (I) show antimicrobial activity and form with a diluent or carrier a pharmaceutical composition which may be administered orally or parenterally.
