Abstract

1309781 Fractionating sperm W SHRIMPTON 9 April 1970 [10 April 1969] 17026/70 Heading B1D [Also in Division C6] Sperm is fractionated into the female-producing X sperm and male-producing Y sperm by cooling a mixture of the sperm and a nutrient medium to a temperature of about 0.8‹C to immobilize the sperm and introducing the cooled mixture into a column of separating medium comprising nutrient medium, to allow separation of the sperm to different levels. A uniform density gradient of separation medium e.g. milk and cream, in the columns 35, Fig. 5, is achieved by combining progressively increasing/decreasing proportions of two liquids of different densities from containers 21 and 22, Fig. 5, by raising/lowering the containers relative to each other by means of a pulley and cable. Mixed liquid of increasing density is introduced into the columns 34 upwardly from the bottom. Further liquid of slightly greater density is then added from container 35. The cooled mixture of sperm and nutrient medium is dispensed from a refrigerated container 47 into spouts 48 and valves 46 are operated to introduce the sperm into the column at the point 44 of average or median density, corresponding to the sperm sample density. X sperm fractions are withdrawn from the bottom of the columns through stopcocks 43. Y sperm fractions may be recovered by draining the entire column or directly, from the top of the column, by means of a pipette. Fractionation efficiencies can be increased by applying a negative pressure of up to 50.8 cm Hg to the columns. Positive pressure may assist the withdrawal of the fractions. Subdued or infra-red light are desirable. According to a further embodiment, Figs. 6 to 8, fractions of the least dense sperm (Y sperm) are withdrawn from the top of column 60 through successively lower apertures 62 into vials 66 by opening valves 64, the central portion is emptied through a single valve 106 and fractions of the densest sperm (X sperm) are withdrawn into vials through successively lower apertures at the bottom of the column. The valves are sequentially operated by actuators through spring-biased cam followers 79 responsive to rotating cams 80 and 82 at the top and bottom of the column, respectively. The column is cooled by a flow of refrigerated air within a chamber 90 generated by impeller 88.
