Abstract

1297581 Cultivating mycobacteria NIPPON OIL CO Ltd 16 Sept 1970 [16 Sept 1969] 44247/70 Heading C6F A method of preparing protein-containing cells involves inoculating a culture medium containing a nitrogen source and a carbon source which comprises carbon dioxide, a saturated hydrocarbon having 1 to 5 carbon atoms or an n-paraffin having 11 to 18 carbon atoms, with Mycobaterium cuneatum ATCC 21498 or Mycobacterium petroleophilum ATCC 21497, culturing the microorganism cells under aerobic conditions, separating the cells from the culture medium and (optionally) freeze-drying or spray-drying them. The carbon source may be a gaseous mixture of carbon dioxide with hydrogen, liquefied petroleum gas consisting mainly of propane and butane, byproduct gases consisting predominantly of hydrogen and methane discharged from naphtha cracking apparatus, fuel gases discharged from petroleum refining apparatus, kerosene, light oil or heavy oil. When using liquid hydrocarbons as the carbon source a small amount of surface active agent may be included in the culture medium and the separated cells after cultivation may be treated with surface active agent and extracted with an organic solvent such as hexane, ethyl ether, acetone, liquefied petroleum gas (butane, propane) or a chlorinating solvent to remove the remaining hydrocarbon sufficiently.
