Abstract

1295570 Heterocyclic substituted salicylic acids MERCK & CO Inc 14 June 1971 [22 June 1970] 27756/71 Heading C2C Novel compounds of Formula I wherein (Het) is a 6-membered ring structure containing up to 3 hetero atoms or a 5-membered ring structure containing up to 4 hetero atoms, the hetero atoms being nitrogen, sulphur and/or oxygen; R 1 is a hydrogen or halogen atom or a lower alkyl, lower alkoxy or halo alkyl radical; and R 2 is a hydrogen or halogen atom are prepared by reaction of a compound of Formula II with carbon dioxide in the presence of an alkalimetal carbonate or where Het is pyrrolyl by action of 2,5-dimethyoxytetrahydrofuran on aminosalicylic acid. Phenols of Formula II above are prepared by demethylation of their methyl ethers of diazotization of the corresponding amines. 4 - ( - m - Methoxyphenyl) - thiazole is prepared by dehydrolhalogenation of the 2-chloro compound prepared by cyclization of α-thio cyanato-m-methoxyacetophenone obtained by action of potassium thiocyanate on m-methoxyphenacyl bromide. 2 - (m - Aminophenyl) - oxazole is prepared by reduction of the nitro compound obtained by cyclization of m-(di-ethoxy ethylaminomethyl) nitrobenzene prepared by condensation of mnitrobenzaldehyde and diethoxyethylamine. 3 - (2 - Thiazolyl) - anisole is prepared by condensation of diethoxyethyl bromide and 3- methoxybenzalthioamide obtained by reaction of thioacetamide and m-cyanoanisole obtained by methylation of the phenol. Pharmaceutical compositions in conventional forms for oral or parenteral administration and having anti-inflammatory, anti-pyretic, analgesic, diuretic, hypoglycemic and anti-fibrinolytic activity comprise an above novel compound and a carrier or diluent.
