1278166 Pyrrolidine and piperidine derivatives YOSHITOMI PHARMACEUTICAL INDUSTRIES Ltd 14 April 1970 17766/70 Heading C2C Novel pyrrolidine and piperidine derivatives of the general formula wherein R<SP>1</SP> is a hydrogen atom or a C 1-3 alkyl group, R<SP>2</SP> is a hydrogen atom or a carbamoyl or C 2-4 alkoxycarbonyl group, NR<SP>3</SP>R<SP>4</SP> is a di-C 1-4 alkylamino, pyrrolidino, piperidino, morpholino or N-methyl-piperazino group, A<SP>1</SP> and A<SP>2</SP> are each a group, the sum of carbon atoms of A<SP>1</SP> and A<SP>2</SP> being 3 or 4, n is 1, 2 or 3 and each X is a halogen atom or a hydroxyl, methyl, methoxy, trifluoromethyl or nitro group or two Xs together form -O-CH 2 -O-, and pharmaceutically acceptable acid addition salts thereof are prepared (a) by reaction of a halide and a pyrrolidine or piperidine derivative of the respective general formulae wherein Y is a halogen atom and (b) by reaction of a benzene derivative of the general formula with a carboxylic acid of the general formula or a reactive derivative thereof, in the presence of a condensing agent, followed optionally by salification of the product. Pharmaceutical compositions having hypocholesterolemic activity comprise, as active ingredient, a pyrrolidine or piperidine derivative of the first general formula above or a pharmaceutically acceptable acid addition salt thereof, together with a pharmaceutically acceptable inert carrier, and may be administered orally.
