Abstract

1,250,394. 3 - Cyclopentyloxy - #<SP>1,3,5(10)</SP>- steroids. AMERICAN HOME PRODUCTS CORP. 23 Jan., 1969 [23 Jan., 1968 ; 24 Oct., 1968], No. 3867/69. Heading C2U. Novel steroids of the formula and their racemates wherein X is (in which R<SP>1</SP> is CH 3 or C 2 H 5 , Z is CO or CH- (#-OH), R is C 1-8 alkyl, ethynyl or chlorethynyl, and R<SP>2</SP> is C 1-8 alkanoyloxy) are prepared (1) by reduction of steroids of the formula in which the #<SP>14</SP> double bond is optional to give, depending on the conditions, a #<SP>1,3,5(10)-8</SP>- tetraener, a #<SP>1,3,5(10)</SP>-triene or an 8-iso- #<SP>1,3,5(10)</SP>-triener; or (2) by etherification with an agent containing a cyclopentyl group of a steroid of the formula wherein X<SP>1</SP> is as X above or is and, if desired or required, oxidizing a resulting 17-ol to a 17-one, reducing a 17-one to a 17-ol with or without concomitant introduction of a 17α-R group, enolacylating a 17-one to a #<SP>16</SP>-17- alkanoyloxy compound, or oxidizing a #<SP>16</SP>-17- alkanoyloxy compound to a 16α-hydroxy-17- ketone or a 16α,17-diol. In these procedures a 17-keto group or a 16- or 17-OH group may be initially protected and subsequently released. These processes may be effected on racemic steroids to give racemic products which may subsequently be resolved, or on optically active steroids. Steroids of the Formula (I) where X has the structure (A), (C), (E) or (F) have estrogenic activity and may be made up into pharmaceutical compositions with suitable carriers.
