Abstract
1,247,449. Determination of lipase; fat or fatty oil emulsion. MERCK. PATENT G.m.b.H. 8 Sept., 1969 [20 Sept., 1968; 27 Aug., 1969], No. 44330/69. Headings B1V and B1X. An agent for the determination of lipase in body fluids comprises an aqueous emulsion of a fat or fatty oil and a pH colour indicator which is one or .more of m-cresol purple, p-xylenolsulphophthalein, dibromothymolsulphophthalein, thymolsulphophthalein, alphanaphtholphthalein, which indicator may be present as 0.001 % to 1 % of the emulsion, optionally together with another colour indicator such as diphenol purple or a coloured compound such as carotin, in concentration up to 0.05%. The fat or oil may be natural or synthetic esters of fatty acids, e.g. tri- or di-glycerides of palmitic, stearic, oleic, linoleic, lauric acids, coconut fat, olive oil (preferred), hydrogenated rape seed oil. Optional components of the emulsion are 0.5 to 30% stabilizers, e.g. gum arabic, polyvinylpyrrolidone; up to 20% emulsifiers, e.g. polyethylene glycol fatty alcohol or alkylphenol ethers, polyoxyethylene derivatives of sorbitol anhydrides, sulphonic acid salts or sulphuric acid esters; up to 2% stabiliser and activator components, e.g. salts of bile acids such as alkali metal cholates, desoxycholates, taurocholates, glycocholates; up to 3% gelling agents, e.g., montmorillonite derivatives; up to 1 % viscosity - increasing agents, e.g. agar, anti-oxidants e.g. mercaptoethanol, cysteine, alphatocopherol acetate, ascorbyl palmitate; up to 1 % biostatic agents, e.g. organo-mercury compounds, p hydroxybenzoic acid esters, hexamethylene tetramine, iodacetates, azides. The indicator may be added as a solution to the emulsion or the solution and emulsion may be stored separately. Unit amounts of emulsion may be stored in synthetic resin squarebased cuvettes with a protective polymeric coating over the surface of the emulsion. In use, the agent is incubated with the sample and the colour developed is compared with a calibrated standard. Pathogenic lipase concentrations may be detected by comparison with results on normal body fluid samples.
