The compartments of a disposable culture assembly 10, Fig. 1, made in polystyrene, are filled with culture media suitable for the primary isolation and differential analysis of bacteria, respectively. Anaerobic culturing conditions are produced by applying the probe device 20, Figs. 1 and 3, down into the surface of the culture medium. The mating surface 22 of the device may be planar (e.g. a thin plastics or glass film or slide) or concave instead of convex, as illustrated, and a multiplicity of times 24 may be fitted. The device 20 may be put up in a sterile heat-sealed plastic bag together with a dropper and a spreader Fig. 6 (not shown). The organisms Proteus mirabilis and E. coli, and several nutrient agar media are mentioned. The container and probe device 20 can be elliptical or trapezoidal instead of rectangular.
