Abstract

1,240,648. N-Substituted isatins. JOHN WYETH & BRO. Ltd. 1 Aug., 1968 [16 Aug., 1967], No. 37596/67. Heading C2C. Compositions comprise isatins of the formula and acid addition and quaternary ammonium salts thereof, wherein R<SP>1</SP> is hydrogen, halogen, alkyl, alkoxy, nitro or haloalkyl, and R<SP>2</SP> is hydrogen or substituted or unsubstituted alkyl or aryl, and a pharmaceutically acceptable carrier, which excludes water and the common organic solvents. Isatins of Formula I wherein R<SP>1</SP> is an alkyl, alkoxy or nitro radical and R<SP>2</SP> is hydrogen or optionally substituted alkyl or aryl, N - (N<SP>1</SP> - methylpiperazino - methyl) isatin and N[-N<SP>1</SP> - (# - hydroxyethyl) piperazino - methyl] isatin, and their acid addition or quaternary ammonium salts are claimed per se, and are prepared by (a) reacting an isatin of the general formula with formaldehyde and a piperazine of the general formula or (b) reacting an isatin of Formula II in the form of an alkali metal salt, with an N,N<SP>1</SP>-disubstituted piperazine of the formula wherein X is halogen or an alkyl- or arylsulphenyl radical, and if desired, converting a free base to the acid addition or quaternary ammonium salt thereof. Examples relate to N - (N<SP>1</SP> - phenylpiperazinomethyl) isatin, N- (N<SP>1</SP> - methylpiperazinomethyl) isatin and N- [N<SP>1</SP> - (# - hydroxyethyl) piperazinomethyl] isatin. The pharmaceutical compositions containing compounds of Formula I and a pharmaceutically acceptable carrier, excluding water and common organic solvents, have hypotensive, CNS or anti-inflammatory activity and may be administered orally, topically or parenterally.
