Glucagon is prepared by condensing H-thr- (tBu) - ser(tBu) - asp(OtBu) - tyr(tBu) - ser- (tBu) - lys(BOC) - tyr(tBu) - leu - asp(OtBu)- ser(tBu) - arg(HX) - arg(HX) - ala - glu - asp- (OtBu) - phe - val - glu - try - leu - net - asnthr(tBu)-OtBu with an excess of AdOC-his- (AdOC) - ser(tBu) - glu - gly - thr(tBu) - phe- OH in which AdOC is adamantyloxy carbonyl and HX is HCl or HBr to give an intermediate AdOC - his(AOC) - ser(tBu) - glu - gly - thr(tBu)- phe - thr(tBu) - ser(tBu) - asp(OtBu) - tyr(tBu)- ser(tBu) - lys(BOC) - tyr(tBu) - leu - asp(OtBu)- ser(tBu) - arg - arg - ala - glu - asp(OtBu) - pheval - glu - try - leu - met - asn - thr(tBu) - OtBu which is claimed per se followed by treatment with trifluoroacetic acid.
