Abstract

1,194,207. Determining creative phosphokinase. SIGMA CHEMICAL CO. 17 July, 1967 [26 Sept., 1966], No. 32839/67. Heading D1X. Creatine phosphokinase (CPK) is determined by reacting ATP with creatine in a test solution containing an unknown quantity of CPK to form ADP, by a method comprising adding to the test solution a sulfhydryl compound, a buffer to maintain a pH of about 9, phospho (enol) pyruvate, pyruvate kinase, lactic dehydrogenase and DPNH, taking a first spectrophotometric measurement about 5 minutes after all the reactants have been added to the test solution, taking a second spectrophotometric measurament at the same wavelength at a known interval of time after the first measurement, and determining the CPK in the test solution from the difference between the first and second measurements. The sulfhydryl compound may be glutathione in its reduced form. The reactions used in the test are Reference has been directed by the Comptroller to Specification 1,153,273.
