Abstract
1,169,807. p-Chlorophenylalanine derivatives. CHAS. PFIZER & CO. Inc. 3 April, 1967 [7 Feb., 1967], No. 15233/67. Addition to 1,153,792. Heading C2C. Novel p-chlorophenylalanine derivatives of the general formula wherein R is hydrogen or C 1 -C 5 alkyl; R<SP>1</SP> is hydrogen or C 1 -C 5 alkanoyl; R<SP>2</SP> is trifluoromethyl or C 1 -C 4 alkyl; and pharmaceutically acceptable acid addition salts thereof are prepared by reaction of a p-chlorobenzyl carbonyl derivative of the general formula with ammonium carbonate and potassium cyanide and treatment of the resulting 5-(pchlorobenzyl)-hydantoin of the general formula with aqueous sodium hydroxide and then glacial acetic acid to give a p-chlorophenylalanine derivative of the general formula followed by optional esterification, conversion to a corresponding N-alkylcarbonyl derivative with an acid anhydride or formation of a pharmaceutically acceptable acid addition salt of the product. Pharmaceutical compositions, useful for lowering the 5-hydroxy-tryptamine concentration in the body, comprise as active ingredient a pchlorophenylalanine derivative of the first general formula above or a pharmaceutically acceptable acid addition salt thereof and a pharmaceutical carrier, and may be administered orally or parenterally.
